RESUMEN

Broad-spectrum antimicrobial doxycycline acts as an inhibitor of protein synthesis and it is widely used in the clinical treatment of various infections by microorganisms that are sensitive to the drug, as well as in animal feed. Its liposolubility guarantees its high tissue bioavailability, being associated with several biochemical changes in the organism and potentially adverse effects on reproduction. This study aims to evaluate the effects of the action of doxycycline on spermatogenesis to provide a complete analysis of the tubular and interstitial compartments and to identify possible changes in the testicular parenchyma. Adult male Wistar rats were divided into three groups: one control (water), and two treated with doxycycline at the doses of 10mg/kg and 30mg/kg, for 30 days. After euthanasia and sample processing, the following parameters were evaluated: a) tubular diameter and height of the seminiferous epithelium; b) volumetric proportions (%) and volumes (mL) of the components of the testicular parenchyma; c) counting testicular germ cell populations; d) evaluation of cell viability. The results of the comparative evaluation between the experimental groups demonstrated a significant increase in the diameter and area of the tubular lumen and a reduction in the count of spermatogonia in the experimental group that received doxycycline hyclate at a dose of 30mg/kg. In the same experimental group, an increase in the overall yield of spermatogenesis was found as a consequence of the increase in the mitotic index.

Tissue changes during embryonic and postnatal development are critical for the success of physiological processes later in mammalian life. Dietary transition from milk to a variety of other food items is one of the factors inducing these changes in mammal species. Blood is utilized as food by only three species of vampire bats among all living mammals. Considering its high protein content, this unique diet is known to induce several metabolic changes, including fasting susceptibility. However, changes in the renal function to meet the excretory needs associated to the blood diet are unknown. Here we aimed at investigating morphological alterations in vampires' kidneys during embryonic and post-natal development in order to better understand the evolutionary adaptations allowing sanguivory. Common vampire bats (Desmodus rotundus) were captured and had their kidneys removed for histological, morphometrical and stereological analysis. Our results showed increased glomerular area and higher glomerular and uriniferous tubules volumetric densities in adults compared to developing bats. These results, together with a higher Renal Somatic Index and a thicker inner medulla also reported for adults, support renal hypertrophy due to increased renal function in blood-feeding vampires as compared to the earlier life stages. We also report a lower foot process density and its different arrange inside the glomerular capsule in adults, indicating an adaptation to a larger extracellular volume formed by increased glomerular filtration. Taken together, kidney morphological changes reported here for vampire bats may reflect in adults' adaptations to a monotrophic strategy.

Many studies have been trying to establish standard protocols for animal experimentation, especially for animal species or strains, to master research variables with high precision. The main mouse strains used in the field of the biology of reproduction are Swiss, Balb/c, and C57BL/6. Since some of the strains show reproduction limitations, such as the size of the litter, the present study aimed to compare their spermatogenic processes to verify differences regarding the testicular parenchyma and germ cell populations, which could explain low offspring production. In addition, the present study provides additional information concerning the testicular parenchyma of such strains, which consequently would help researchers to choose the most suitable strain for reproductive studies. Six adult male mice were used for each of the strains. After euthanasia, the testes were weighed, fixated with Karnovsky fixative, embedded in methacrylate, sectioned, and stained with toluidine blue/sodium borate 1%. Morphometrical analyses from the testicular parenchyma (seminiferous tubules and interstitium) were made using the software ImageJ. Germ and Sertoli cells populations were counted in seminiferous tubules cross-sections at stage I of the seminiferous epithelium cycle. The lowest body and testicular weights were observed in C57BL/6 animals, followed by Balb/c and Swiss, however, the relative testes, parenchyma, and albuginea weights were significantly lower only in C57BL/6. Despite the seminiferous tubules and seminiferous epithelium proportions were lower in Swiss animals, their relative amount related to the body weight was the same among strains. The total number of germ cells was higher in Swiss animals, reflecting higher spermatogenic yield and daily sperm production. Due to the lower relative number of Sertoli cells, the Swiss animals showed the highest Sertoli cell index and support capacity. On the other hand, the lowest pathological indexes regarding the germ cells were observed in Balb/c animals, followed by Swiss and C57BL/6. In the interstitium, the proportion of blood vessels was lower in Swiss mice, while the lymphatic cell proportion was lower in C57BL/6 animals. Moreover, the highest proportions of Leydig cells and macrophages were noticed in Swiss mice, which may indicate increased testosterone levels. Altogether, such observations must be taken into account when choosing any of the studied strains for reproduction studies.

ETHNOPHARMACOLOGICAL RELEVANCE: Roots of Pfaffia glomerata are used in traditional medicine as aphrodisiacs and sexual stimulants. AIM OF THE STUDY: The aim of this study was to evaluate the action of the hydroalcoholic extract from the roots of Pfaffia glomerata on the Leydig cells, cavernous bodies and other penile constituents, as well as on serum testosterone and 17ß-estradiol levels of adult mice. MATERIALS AND METHODS: Mature male Swiss mice were divided into 6 groups: control (water), sildenafil citrate, 3 groups receiving daily doses of P. glomerata extract (100, 200 and 400 mg/kg) and one group receiving intermittent doses of P. glomerata (200 mg/kg/3-3d). RESULTS: The proportions of blood vessels, lymphatic space and estradiol levels were increased. On the other hand, reduction of testosterone levels due to Leydig cells death was observed. As for penile parameters, volumetric proportions of cavernous bodies, collagen and nitric oxide were increased, while smooth muscle content was decreased. CONCLUSIONS: Despite that the long term intake of P. glomerata extract was related to a stimulant action, reduction on Leydig cell viability induced decreased testosterone production.

The seminiferous epithelium goes through multiple changes which enables the differentiation of a spermatogonia in a fully mature spermatozoon. The timing of these changes is species-specific and influences the duration of the reproductive cycles. Bats are among wild mammals whose coordination between male and female reproductive cycles are imperative, since most females show seasonal preferences, even in the Tropics. This seasonal variation demands constant sperm production ready for spermiation in order to guarantee its genetic dispersion and reproduction success. Despite their abundance, little is known about the duration of reproductive cycles in Neotropical bat species, a relevant information for the species management and for conservational strategies regarding anthropogenic and climate influences on bats reproduction. In this study, we aimed at characterizing the stages of the seminiferous epithelium cycle (SEC) of the fruit bat Artibeus lituratus and to determine its duration based on the immunohistochemical analysis of the bromodeoxyuridine (BrDU) activity. SEC stages were characterized according to the tubular morphology method and the frequency of each stage was estimated. After intratesticular injections of BrDU, the animals were euthanized at different times, and the estimation of SEC duration was performed by observing the most advanced germ cells in the seminiferous epithelium. The most advanced stained cells after 2 days of BrdU injection were the primary spermatocytes in pachytene, transitioning from stages 1-2 of the SEC. Within 2 days, we found a progression of 30.42% of the SEC, and an entire cycle lasted 6.58 days on average. Considering that 4.5 seminiferous epithelium cycles are necessary for the whole spermatogenic processes to be completed, the total length of spermatogenesis in A. lituratus was estimated at 29.61 days. Our findings support a pattern of bimodal seasonal polyestry for this species, with rapid spermatogenic cycles.

Diphylla ecaudata is a hematophagous bat endemic of South America, with food preference for bird blood. Given the lack of information about the reproductive activity of this species, this study aimed to describe the testicular morphology and histomorphometry of D. ecaudata in order to understand its reproductive biology, specially spermatogenesis. The animals were collected in Lajes city, Rio Grande do Norte, Brazil. Following euthanasia, the testes were histologically processed for morphological, morphometric, ultrastructural and immunohistochemical analyses. Their average body weight was 24.64g, with a gonadosomatic index of 0.49%, tubulesomatic index of 0.47%, and a total of 32.20m of seminiferous tubules per gram of testis. The pre-meiotic, meiotic, and post-meiotic phases accounted for 56.20%, 9.30%, and 34.50% of the seminiferous epithelium cycle, respectively. The ultrastructure of spermiogenesis was similar to that described in other mammals and the perforatorium was not observed in the sperm. Androgen receptors were detected in Sertoli cell nuclei and Leydig cell cytoplasm, while aromatase enzyme was detected only in Sertoli cell nuclei. FGF2 and BCL-2 activities were detected in the cytoplasm of zygotene and pachytene primary spermatocytes, as well as round and elongated spermatids. D. ecaudata showed testicular pattern similar to other mammals and characteristics common to other bat species. This species stood out for its high efficiency of Sertoli cells, which presented high capacity to support germ cells, besides the highest sperm production rates among those already recorded. This study is the first step towards the knowledge of D. ecaudata reproduction and the first description of its spermatogenesis.

ABSTRACT Objective: To assess the effects of atorvastatin calcium in the treatment of dexamethasone-induced osteoporosis. Methods: Osteoporosis induction consisted of the administration of an intramuscular dose of 7.5 mg/kg of body weight of dexamethasone, once a week for four weeks, except for the control animals (G1). The animals were divided into the following groups: G1 (control group without osteoporosis), G2 (control group with untreated osteoporosis), G3 (control group with osteoporosis treated with sodium alendronate 0.2 mg/kg) and G4 (group with osteoporosis treated with atorvastatin calcium 1.2 mg/kg). Serum alkaline phosphatase, bone alkaline phosphatase, and biometric and bone histomorphometric assessments were performed after 30 and 60 days of treatment onset. Results: In relation to the biometric and histomorphometric analyses, at 60 days of treatment, G4 presented bone density (Seedor index), bone trabecular density, and cortical thickness of 0.222 ± 0.004 g/cm, 59.167 ± 2.401%, and 387,501 ± 8573 µm, respectively, with a positive and statistically significant difference (p < 0.05), in relation to G2. At 30 and 60 days of treatment, G4 presented statistically significant serum levels of alkaline phosphatase alkaline phosphatase (p < 0.05) that were higher than all groups (7.451 ± 0.173 µg/L and 7.473 ± 0.529 µg/L, respectively). Conclusion: Treatment with atorvastatin calcium demonstrated the ability of this drug to increase osteoblastic activity and bone tissue repair activity, acting differently from alendronate sodium, which demonstrated predominantly antirebsorptive activity.

Métodos: A indução da osteoporose consistiu na administração de dexametasona na dose de 7,5 mg/kg de peso corporal, por via intramuscular, uma vez por semana durante quatro semanas, à exceção dos animais do grupo controle (G1). Os animais foram distribuídos nos seguintes grupos: G1 (grupo controle sem osteoporose), G2 (grupo controle com osteoporose sem tratamento), G3 (grupo controle com osteoporose tratado com alendronato de sódio 0,2 mg/kg) e G4 (grupo com osteoporose tratado com atorvastatina cálcica 1,2 mg/kg). Após 30 e 60 dias do início do tratamento dos animais, foram feitas as dosagens dos níveis séricos de fosfatase alcalina, fosfatase alcalina óssea, avaliação biométrica e histomorfométrica óssea. Resultados: Em relação às análises biométricas e histomorfométricas, aos 60 dias de tratamento o G4 apresentou densidade óssea (índice Seedor), densidade trabecular óssea e espessura da cortical de 0,222 ± 0,004 g/cm, 59,167 ± 2,401% e 387,501 ± 8,573 µm, respectivamente, com diferença positiva, estatisticamente significativa (p < 0,05), em relação ao grupo G2. Aos 30 e 60 dias de tratamento, o G4 apresentou níveis séricos de fosfatase alcalina óssea estatisticamente significativos (p < 0,05) e superiores a todos os grupos (7,451 ± 0,173 µg/L e 7,473 ± 0,529 µg/L, respectivamente). Conclusão: O tratamento com atorvastatina cálcica demonstrou a capacidade desse fármaco de aumentar a atividade osteoblástica e a atividade reparadora tecidual óssea, atuar de forma diferente do alendronato de sódio, que demonstrou atividade preponderantemente antirreabsortiva.

Introdução: Além da indução da osteoporose, os glicocorticoides ocasionam aumento da resistência à insulina e gliconeogênese hepática, tendo como consequência a hiperglicemia. Objetivo: Avaliar comparativamente os efeitos do alendronato de sódio e da atorvastatina cálcica nos níveis séricos de glicose e insulina na osteoporose induzida com dexametasona. Métodos: A indução da osteoporose consistiu na administração de dexametasona na dose de 7,5 mg/kg de peso corporal, uma vez por semana durante 4 semanas, à exceção dos animais do grupo controle (G1). Os animais foram distribuídos nos seguintes grupos: G1 (grupo controle sem osteoporose), G2 (controle com osteoporose sem tratamento), G3 (com osteoporose tratado com alendronato de sódio 0,2 mg/kg) e G4 (com osteoporose tratado com atorvastatina cálcica 1,2 mg/kg). No período de 30 e 60 após o início do tratamento, foram coletadas amostras de sangue para as dosagens dos níveis séricos de glicose e insulina. Resultados: Os grupos G2 e G3, quando comparados com o grupo normal G1, apresentaram aumento da glicemia e insulinemia durante todo o período experimental. O grupo G4 apresentou, com 30 dias, aumento da glicemia e insulinemia e, com 60 dias, aumento da glicemia e queda da insulinemia. Conclusão: Os resultados demonstraram o quadro de hiperglicemia consequente do aumento da resistência à insulina, presentes na indução da osteoporose pela dexametasona. O alendronato de sódio não ocasionou nenhuma melhora da glicemia e insulinemia. A atorvastatina cálcica ocasionou agravamento da hiperglicemia e hiperinsulinemia, potencializando o quadro de resistência à insulina e levando a uma insuficiência relativa de insulina característica do diabetes mellitus tipo 2.


Introduction: In addition to the induction of osteoporosis, glucocorticoids cause increased insulin resistance and hepatic gluconeogenesis resulting in hyperglycemia. Objective: Evaluate the effects of sodium alendronate and atorvastatin calcium on serum glucose and insulin levels in osteoporosis induced by dexamethasone. Methods: The induction of osteoporosis consisted of the administration of dexamethasone at a dose of 7.5 mg / kg body weight, once a week for 4 weeks, except for the control animals (G1). The animals were divided into the following groups: G1 (control group without osteoporosis), G2 (control with untreated osteoporosis), G3 (with osteoporosis treated with sodium alendronate 0.2 mg / kg) and G4 (with osteoporosis treated with atorvastatin calcium 1,2 mg / kg). In the 30 and 60 period after the start of the treatment blood samples were collected for dosages of serum glucose and insulin levels. Results: The G2 and G3 groups, when compared with the normal group G1, presented increased glycemia and insulinemia throughout the experimental period. The G4 group presented a 30-day increase in glycemia and insulinemia and at 60 days increased glycemia and decreased insulinemia. Conclusion: The results demonstrated the hyperglycaemia associated with the increase in insulin resistance present in the induction of osteoporosis by dexamethasone. Sodium alendronate did not cause any improvement in glycemia and insulinemia. Atorvastatin calcium caused worsening of hyperglycemia and hyperinsulinemia enhancing insulin resistance, leading to a relative insufficiency of insulin characteristic of type 2 diabetes mellitus.

Among all bat species, Desmodus rotundus stands out as one of the most intriguing due to its exclusively haematophagous feeding habits. However, little is known about their spermatogenic cycle. This study aimed at describing the spermatogenic process of common vampire bats through testicular histomorphometric characterization of adult specimens, spermatogenic production indexes, description of stages of the seminiferous epithelium cycle and estimative of the spermatogenic process duration. Morphometrical and immunohistochemical analyzes for bromodeoxiuridine were conducted under light microscopy and ultrastructural analyzes were performed under transmission electron microscopy. Vampire bats showed higher investment in gonadal tissue (gonadosomatic index of 0.54%) and in seminiferous tubules (tubulesomatic index of 0.49%) when compared to larger mammals. They also showed a high tubular length per gram of testis (34.70 m). Approximately half of the intertubular compartment was found to be comprised by Leydig cells (51.20%), and an average of 23.77x106 of these cells was found per gram of testis. The germline cells showed 16.93% of mitotic index and 2.51% of meiotic index. The overall yield of spermatogenesis was 60% and the testicular spermatic reserve was 71.44x107 spermatozoa per gram of testis. With a total spermatogenesis duration estimated at 37.02 days, vampire bats showed a daily sperm production of 86.80x106 gametes per gram of testis. These findings demonstrate a high sperm production, which is commonly observed in species with promiscuous mating system.

ABSTRACTThe aim of this study was to investigate the effect of the bark extractBathysa cuspidata on chemically induced preneoplastic colorectal lesions in Wistar rats. Forty male rats were randomly divided into four groups (n = 10 each): saline (control group, oral administration of saline solution 0.9%); dimethylsulfoxide (DMSO, vehicle control), B200 (treated with 200 mg/kg bark extract ofB. cuspidata), and B400 (treated with 400 mg/kg bark extract ofB. cuspidata). Administration of treatments was carried out by the gavage. The animals received four subcutaneous injections of 1,2-dimethylhydrazine (DMH, 40 mg/kg) in the initial two weeks of the experiment to induce preneoplastic colorectal lesions. After 15 weeks, the animals were euthanized and the presence of aberrant crypt foci (ACF), body weight, biochemical analyses, and oxidative stress markers were measured. The extract ofB. cuspidata decreased the levels of superoxide dismutase (SOD), but did not influence the levels of catalase (CAT), malondialdehyde (MDA), nitric oxide or protein carbonyl, compared with the saline group. The animals supplemented with a more concentratedB. cuspidata extract (B400) showed a significant reduction in the number of ACF in all the portions of the intestinal mucosa. The study demonstrated that the bark extract ofB. cuspidata at 400 mg/kg reduced the preneoplastic colorectal lesions in an animal model of colon cancer and that the effect could be dose-dependent.

The aim of this study was to investigate the effect of bark extract Bathysa cuspidata (BCE) in the reconstitution of hepatic parenchyma and stroma after dysfunction induced by the CCl4. Liver lesions were induced by intraperitoneal administration of CCl4 (1 mL/kg) every 48 h for 12 days. The animals were treated with B. cuspidata extract (BCE) administered by gavage for another 12 days. Forty-nine rats were randomized into seven treatment groups with seven animals receiving CCl4, BCE (200, or 400 mg/kg) and the vehicle DMSO alone, or in different combinations. The extract alone showed no evidence of hepatic toxicity. In general, rats acutely exposed to CCl4 without the treatment with BCE presented high ALT and AST serum levels and high tissue content of lipid hydroperoxides, malondialdehyde, and lipid droplets. BCE administration, especially at 400 mg/kg attenuated significantly all these parameters. Furthermore, the antioxidant activities of the enzymes superoxide dismutase and catalase were significantly increased in the groups receiving this extract. These results showed that the extract of B. cuspidata stem bark stimulated the antioxidant defense system and reduced the morphological and functional liver damage in Wistar rats previously exposed to CCl4.

INTRODUÇÃO: A população brasileira tem seguido a tendência dos países desenvolvidos, cuja característica é o aumento do consumo de gorduras e sedentarismo. OBJETIVO: Investigar os efeitos da dieta hiperlipídica e do treinamento sobre o peso corporal, o consumo alimentar, a massa corporal magra, os lipídios plasmáticos, o peso do fígado e a aterosclerose em camundongos com deficiência de apolipoproteína E (apoE-/-). MÉTODOS: vinte e seis camundongos foram divididos em grupos/dieta: NS (normolipídica e sedentário), HS (hiperlipídica e sedentário), NT (normolipídica e treinamento) e HT (hiperlipídica e treinamento). Aferiu-se peso corporal (PC), consumo alimentar (CA) e peso relativo do fígado (PRF). Calculou-se a água da carcaça (A%) pela diferença do peso pré e pós-secagem, gordura (G%) por Soxhlet e proteína (P%) por Kjeldahl. Determinaram-se colesterol total (CT), triglicerídeos (TG) e HDL por método enzimático-colorimétrico e LDL e VLDL pela formula de Friedewald. Analisou-se a aterosclerose por fotomicrografia. Utilizou-se ANOVA e o método de Duncan, com P < 0,05. RESULTADOS: No PC inicial e final, não houve diferença de G% e P% entre os grupos (P > 0,05). A dieta hiperlipídica aumentou o CA (365,3 ± 5,09 vs. 340,16 ± 6,32) e LDL (452,20 ± 114,63 vs. 329,60 ± 77,54) nos animais sedentários e o CT (858,75 ± 140,84 vs. 320,87 ± 157,81) e o LDL (839,36±139,94 vs. 282,66±166,92) nos treinados (P < 0,05). Já o treinamento aumentou o CA (350,4 ± 7,81 vs. 340,16 ± 6,32) e A% (64,23 ± 2,46 vs. 62,62 ± 1,47) nos camundongos normolipídicos e diminuiu o CA (336,68 ± 6,23 vs. 365,3 ± 5,09), TG (22,75 ± 12,36 vs. 66,00 ± 21,88), HDL (14,83 ± 3,95 vs. 37,60 ± 13,12), VLDL (4,55 ± 2,47 vs. 13,20 ± 4,37) e aterosclerose (0,35 ± 0,13 vs. 0,56 ± 0,12) nos hiperlipídicos (P < 0,05). CONCLUSÃO: A dieta hiperlipídica pode desencadear distúrbios no metabolismo lipídico e, com isso, desencadear doenças cardiovasculares; porém, associada ao treinamento, pode diminuir a aterosclerose.


INTRODUCTION: The Brazilian population has followed the trend of developed countries, whose characteristic is the increased consumption of fats and sedentary lifestyle. OBJECTIVE: To investigate the effects of high fat diet and training on body weight, food intake, lean body mass, plasma lipids, liver weight and atherosclerosis in mice with apolipoprotein E (apoE-/-) deficiency. METHODS: 26 rats were divided into groups/diet: NS (normolipidic and sedentary), HS (high fat and sedentary), NT (normolipidic and training) and HT (high fat and training). Body weight (BW), food intake (FI) and relative liver weight (RLW) were measured in a Gehaka BG2000 scales. The water from the carcass (A%) was calculated from the weight difference between the pre-and post-drying ,the fat (% F) was calculated by Soxhlet, and the protein (P%) by Kjeldahl . We determined total cholesterol (TC), triglycerides (TG) and HDL by colorimetric enzymatic method and VLDL and LDL by the Friedewald formula. Atherosclerosis was analyzed by photomicrography. We used ANOVA as well as the Duncan's method, P<0.05. RESULTS: In the initial and final BW, there was no difference in F% and P% between groups (P>0.05). The high fat diet increased the FI (365,3±5,09 vs. 340,16±6,32) and LDL (452,20±114,63 vs. 329,60±77,54) in the sedentary animals and TC (858,75±140,84 vs. 320,87±157,81) and LDL (839,36±139,94 vs. 282,66±166,92) in trained ones (P<0.05). On the other hand, training increased the CA (350,4±7,81 vs. 340,16±6,32) and A% (64,23±2,46 vs. 62,62±1,47) in normolipidic mice and decreased FI (336,68±6,23 vs 365,3 ± 5,09), TG (22,75±12,36 vs. 66,00±21,88), HDL (14,83±3,95 vs. 37,60±13,12), VLDL (4,55±2,47 vs. 13,20±4,37) and atherosclerosis (0,35±0,13 vs. 0,56±0,12) in hyperlipidic mice (P<0.05). CONCLUSION: A high fat diet can trigger disturbances in the lipid metabolism and thereby trigger cardiovascular disease, but when associated with activity may decrease atherosclerosis.

Se controlaron peso corporal (PC), consumo de alimentos (CA) y peso relativo del hígado (PRH). Desde otro punto de vista, el entrenamiento aumentó el CA (350,4 ± 7,81 vs. 340,16 ± 6,32) y A% (64,23 ± 2,46 vs. 62,62 ± 1,47) en los ratones normolipídicos y disminuyó el CA (336,68 ± 6,23 vs. 365,3 ± 5,09), TG (22,75 ± 12,36 vs. 66,00 ± 21,88), HDL (14,83 ± 3,95 vs. 37,60 ± 13,12), VLDL (4,55 ± 2,47 vs. 13,20 ± 4,37) y la aterosclerosis (0,35 ± 0,13 vs. 0,56 ± 0,12) en los hiperlipídicos (P < 0,05). CONCLUSIÓN: La dieta hiperlipídica puede causar disturbios en el metabolismo lipídico y, con eso, producir enfermedades cardiovasculares; no obstante, si está asociada con el entrenamiento, puede reducir la aterosclerosis.

RESUMEN

This study was undertaken to determine whether colon cancer (CC) and chronic swimming exercise alter rat testis. Eleven weeks old rats were distributed into control group (n=6) and the groups that were induced to develop CC by dimethylhydrazine injections (nEG, EG0, EG2 and EG4; n=10 each group). In the group nEG, the rats did not swim, whereas groups EG0, EG2 and EG4, underwent a swimming program with distinct loads (0, 2 and 4% of body mass, respectively) for 35 weeks. The morphometry, stereology and cell counts showed damage caused by the CC on the germ epithelium. These results were noteworthy since this was the first report to associate the CC with testicular damage. Swimming exercise had no significant role in reducing, or increasing the CC effects on the testis, despite having slightly improved the testis structure of the exercised rats without load. In conclusion, CC caused testis impairment, which could not be avoided by the swimming exercise.

Although some animal models of food allergy in have already have been described, none of them uses the allergen in the animals' diet. This work describes the comparison between two developed models of food allergy in BALB/c mice, based in the administration of the allergen in the diet or by intragastric way. The experiment last for 28 days and the animals had been sensitized by means of subcutaneous injection in 1st and 14th days with in natura extract milk, bovine extract meat or frog extract meat. The experimental model that uses the allergen in the unbroken form presented morphometric alterations when compared with the one that used the heat treat allergen. It was noticed the existence of some more resistant proteins than others related to the denaturation, once compared the results of the two models; the differences had been more prominent for the milk and frog allergens. These results confirm the epidemiologic data of allergy incidence in the world's population (AU)


Aunque algunos modelos animales para estudio in vivo de alergia alimentaria hayan sido descriptos, ninguno de ellos utiliza el alergeno en la dieta de los animales. Este trabajo describe la comparación entre dos modelos experimentales de alergia alimentaria desarrollados en los ratones BALB/c, inducida por la administración del alergeno en la dieta o por la vía intragastrica. El experimento fue desarrollado por un período de 28 días y los animales fueron sensibilizados por inyección subcutánea en el 1º y 14º días con extracto de leche in natura, extracto de carne de buey o extracto de carne de rana. El modelo experimental que recibió el alergeno intacto presentó las alteraciones morfométricas más evidentes cuando fueron comparadas con los que recibió el alergeno tratado térmicamente. Se evidenció la presencia de proteínas más resistentes que otras en lo que se refiere a la desnaturación, una vez que cuando fueron comparados los dos modelos, las diferencias fueron más claras para los alergenos de la leche y de la carne de rana. Estos resultados confirman los datos epidemiologicos de incidencia de alergia en la población mundial (AU)

RESUMEN

The occurrence of seasonality in bullfrog (Lithobates catesbeianus) spermatogenesis was analyzed by quantifying germ cell populations throughout the year, which were divided into bimesters, from November 2001 to October 2002. Body weight, liver weight, fat body weight and gonadal weight were taken and bimonthly grouped. Using these data, the gonadosomatic, hepatosomatic, and liposomatic indexes were calculated. All animals were kept in the same cement cages in a frog farm, and sampled every month. The cages were underthe influence of climate variables changes, such as photoperiod, temperature, rainfall and relative humidity.Body weight was higher during Spring and Summer, and a significant reduction has occurred between Julyand August. The highest gonadosomatic index values were registered from May to October, coinciding withthe period of gonadal preparation for reproduction. Also, the reproduction of bullfrog is directly related toenvironmental factors, mainly due to Summer and Winter humidity variations.

FUNDAMENTO: A aterosclerose é uma doença inflamatória crônica de origem multifatorial que ocorre em resposta à agressão endotelial. O fungo Monascus ruber apresenta atividade hipocolesterolêmica e polifenóis presentes no resíduo de café apresentam atividade antioxidante, podendo auxiliar na prevenção de doenças cardiovasculares. O resíduo de café possui quantidade significativa de açúcares fermentescíveis, constituindo-se em substrato apropriado para o cultivo de fungos. OBJETIVO: O objetivo deste estudo foi avaliar o efeito dos resíduos de café seco e fermentado por Monascus ruber no metabolismo lipídico de camundongos knockout Apo E. MÉTODOS: O ensaio biológico foi realizado com 30 camundongos knockout para o gene Apo E, divididos em cinco grupos e submetidos a diferentes tratamentos. Foi realizada a prospecção fitoquímica e quantificação de compostos fenólicos dos resíduos fermentado e sem fermentar. O soro dos animais foi analisado utilizando kits enzimáticos e o tecido aórtico incluso em parafina e corado com H/E para realização da análise histopatológica. RESULTADOS: O resíduo de café sem fermentar 2%, em relação ao grupo controle, diminuiu em 42% o nível sérico de triacilgliceróis e em aproximadamente 41% a fração VLDL-c. Os grupos dos animais alimentados com 10% de resíduo não fermentado e 2% de resíduo fermentado diminuíram a área de lesão 10,5% e 15,4%, respectivamente, quando comparados ao controle. O resíduo fermentado apresentou um teor de compostos fenólicos superior ao resíduo não fermentado. CONCLUSÃO: O presente estudo mostra que a fermentação do resíduo de café apresenta potencial efeito benéfico sobre as doenças cardiovasculares, especialmente a aterosclerose.

METHODS: The biological assay was performed with 30 ApoE knockout mice, divided into five groups and undergoing different treatments. The phytochemical prospection and quantification of phenolic compounds of the fermented and non-fermented coffee residues were performed. The sera of the animals were analyzed by using enzyme kits, and the aortic tissue was embedded in paraffin and stained with hematoxylin and eosin to undergo histopathological analysis. RESULTS: Comparing with the control group, the group receiving 2% non-fermented coffee residue showed a reduction of 42% in the serum levels of triacylglycerols and of approximately 41% in VLDL-c. The groups receiving 10% non-fermented coffee residue and 2% fermented coffee residue showed reductions in the lesion areas of 10.5% and 15.4%, respectively, as compared with the control group. The fermented coffee residue showed a higher content of phenolic compounds as compared with the non-fermented coffee residue. CONCLUSION: The present study showed that coffee residue fermentation has a potentially beneficial effect on cardiovascular diseases, especially atherosclerosis.

BACKGROUND: Cardiovascular diseases are the main causes of death in the Western world and are manifested by atherosclerosis. Depending on its intensity, regular aerobic exercise may be either beneficial or harmful to the atherosclerosis process. AIM: The aim of this study was to verify the effects of aerobic exercise training of different intensities on the profile of atherosclerotic lesions and serum lipid, and in the hepatic oxidative balance of low-density lipoprotein receptor-deficient (LDLr(-/-)) mice previously developed with atherosclerosis. METHODS: All animals were submitted to a three-month high-fat and high-cholesterol diet regime. The animals were then randomly divided into no exercise (G1, n=9), low-intensity aerobic exercise (G2, n=10, 8 weeks of treadmill running, 30 min/day(-1) at 8-10 m/min(-1)) and moderate-intensity aerobic exercise (G3, n=10, 8 weeks of treadmill running, 30 min/day(-1) at 10-16 m/min(-1)) groups. Serum total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C) and triglycerides (TG), and oxidative damage (protein carbonyls and lipid hydroperoxides) were measured. The activity of catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GPx) in the liver tissue was assessed. RESULTS: G2 (0.015 ± 0.005cm(2)) and G3 (0.014 ± 0.001cm(2)) presented lower aortic fat deposition than G1 (0.039 ± 0.005cm(2)). G2 and G3 exhibited higher HDL-C, TG and CAT activity, but lower lipid peroxidation and carbonyl protein than G1. SOD values were higher in G3 than G2 and G1, and GPx was higher in G2 than in G3 and G1. CONCLUSIONS: Our protocols of low- and moderate-intensity aerobic exercise training (30 min daily for 8 weeks) induced similar benefits in LDLr(-/-) mice with atherosclerosis.
